GENEPOP analysis We will analyse the full dataset of 11 allozyme loci in the two mosquito populations using the GENEPOP population genetics software. Run the analyses under the following GENEPOP options: 1. Hardy Weinberg Exact Tests 2. Linkage Disequilibrium 3. Population Differentiation 5. Basic Information, Fis and gene diversities 6. Fst & other correlations Overall you need to: 1. Summarise allele frequencies at all loci and in both populations. 2. Calculate the number of polymorphic loci and allelic diversities for each population 3. Calculate Ho and He for each locus and the average values for populations 4. Test for departures from Hardy-Weinberg equilibrium in each population and overall. 5. Calculate F statistics. 6. Test for population differentiation 7. Test for pairwise disequilibria between polymorphic loci within populations. Write up On the basis of your GENEPOP analysis, you should write a maximum of two-sides (11pt font (Times New Roman, Arial, Calibri); 1.5 line spacing), summarising the results of the analyses and describing the inferences (in terms of evolutionary forces/demographic processes) that can be made about the populations from the genetic data. Your conclusions should be supported by quoting the relevant parameters, with statistical tests. These can be in tables or figures in addition to the 2 pages of text. Don't just cut and paste the GENEPOP results directly – they may need to be summarised, edited or formatted to make appropriate, useable tables. You should aim for a style similar to the results and discussion section of a population genetics paper describing genetic diversity and population structure, such as those in Molecular Ecology or Conservation Genetics. i.e. we'll be going through the same process as if we were a conservation geneticist analysing and writing up the data for real.
